Gli afidi rappresentano un gruppo di insetti fitomizi di primaria importanza in agricoltura. Myzus persicae è considerato l'afide più dannoso a livello mondiale e uno degli insetti che ha sviluppato il maggior numero di resistenze agli insetticidi. Durante il mio dottorato ho analizzato alcuni geni correlati alla fitness di M. persicae con l'obbiettivo di fornire nuovi target molecolari per la gestione di questa specie in agricoltura. Gli afidi possiedono cromosomi olocentrici in grado di mantenere variazioni di numero e struttura. Abbiamo raccolto in una review tutti i dati citogenetici disponibili per M. persicae evidenziando che la variazione più comune è la traslocazione cromosomica reciproca tra gli autosomi 1 e 3, correlata alla fitness, in quanto conferisce la resistanza a organofosfati e carbammati. Gran parte dei cloni di M. persicae con la tA1-A3 sono di colore rosso e vivono su tabacco: con l'obbiettivo di studiare la relazione tra colore e struttura cromosomica abbiamo mappato i geni per la sintesi dei carotenoidi in M. persicae, Acyrthosiphon pisum e Neotoxoptera formosana localizzandoli sull'autosoma 1. M. persicae e A. pisum mostrano sintenia, condividendo un frammento di 246,000pb, nel quale abbiamo individuato siti fragili che potrebbero aver favorito la tA1-A3, in presenza di nicotina. Per misurare la fitness degli afidi sono stati impiegati numerosi indici e approcci e questo ha reso complicato confrontare dati provenienti da autori diversi. Ho sviluppato uno strumento bioinformatico in Python (Afit-tool) e un modello Agent Based in NETLOGO (AFIT) per standardizzare e rendere più semplice l'analisi della fitness. Ho testato Afit-tool con 6 cloni di M. persicae per verificare l'effetto delle variazioni del cariotipo sulla fecondità, scoprendo che l'instabilità cariotipica non riduce la fitness e che al contrario i cloni con la tA1-A3 hanno una fitness più alta. Abbiamo usato Afit-tool anche per confrontare 4 cloni di M. persicae con diversa sensibilità ai piretroidi e con differenti alleli kdr scoprendo che mutazioni in questo locus hanno un peso sulla fitness. Gli afidi producono una saliva ricca di enzimi che interagiscono con le difese della pianta. Un esempio unico di questo sono le trealasi. Ho indagato la storia evolutiva delle trealasi (treh) scoprendo che gli afidi hanno il più alto numero di duplicazioni del gene treh. Gli Emitteri hanno duplicazioni specie-specifiche e le regioni genomiche in cui si trovano i geni treh hanno livelli di sintenia molto più bassi rispetto agli altri insetti a causa dell'olocentrismo. Le isoforme mostrano invece negli Emitteri la più alta diversità suggerendo eventi di subfunzionalizzazione. Le trealasi negli afidi potrebbero agire da soppressori del pathway di difesa PAD4 e le piante potrebbero aver evoluto degli inibitori. Questa “corsa agli armamenti” potrebbe essere la causa della grande radiazione della famiglia genica treh individuata negli afidi. Il ruolo delle heat shock proteins (hsp) nella plasticità fenotipica non è mai stato studiato negli afidi, per cui ho caratterizzato il gene hsp90 in 17 cloni di M. persicae con diverse strategie riproduttive e produzione di forme alate e scoperto che mutazioni che alterano la proteina sono presenti solo in cloni anolociclici. Questo suggerisce che hsp90 potrebbe compensare l'effetto negativo di altre mutazioni che impediscono la partenogenesi ciclica. Il gene foraging (for) è invece un esempio notevole di un singolo gene responsabile di caratteri legati al comportamento e alla plasticità fenotipica; abbiamo caratterizzato una parte del gene for in 12 cloni di M. persicae scoprendo che la produzione di forme alate, in grado di colonizzare l'ambiente più rapidamente, sembra essere correlata alla funzionalità di questo gene.

Abstract
Aphids are phloem sap sucking insects of primary agricultural importance. The polyphagous Myzus persicae is the most important aphid pest and one of the most widely pesticide resistant insects. During my PhD, I investigated diverse molecular traits influencing M. persicae fitness, with the aim to identify new molecular targets for the field management of this species. We reviewed data attesting the presence of such changes in M. persicae evidencing that the most common variant consists of a translocation between autosomes 1 and 3 (tA1-A3), linked to fitness, since it confers organophospate and carbamate resistance. All M. persicae clones with the tA1-A3 are red morphs so, to verify the relationship between colour and chromosome structure, we studied the chromosomal localization of genes involved in carotenoids biosynthesis in M. persicae, Acyrthosiphon pisum and Neotoxoptera formosana. M. persicae and A. pisum genes were in a syntenic 246,000 bp long fragment on autosome 1 in all 3 species. We investigated and found fragile sites that, in presence of nicotine, could favour the tA1-A3 observed in the red M. persicae tobacco feeding clones. Many indices have been used to measure aphid fitness and data were collected in diverse contexts, making complex the comparison of the published data. I set up a Python bioinformatics tool (Afit-tool) and a NETLOGO AB Model (AFIT) to help researchers to easily calculate fitness indices. We tested Afit-tool with 6 M. persicae clones to investigate the effect of karyotype rearrangements on fitness. Our data demonstrated that there are no fitness costs considering karyotype variations but rather M. persicae possessing the tA1-A3 has higher fitness than a M. persicae lineage with standard karyotype, thus evidencing the potentiality of holocentrism to reduce chromosome rearrangements effects. We also used Afit-tool to calculate fitness of 4 M. persicae lineages with diverse pyrethroid sensitivity and kdr alleles. The susceptible M. persicae lineage achieved the highest fitness scores suggesting a direct link between kdr mutations and fitness. Aphids secrete enzyme-rich saliva, which interacts with plant defences. An interesting example of such enzyme is trehalase (treh). We have inferred the evolutionary history of aphid trehalases and tested their functional divergence, combining nucleotides sequences phylogeny, protein conserved domains and genomic synteny analysis. Aphids show the highest number of treh gene duplications and possess duplicated acid trehalase (ath) genes as well. Most of Hemiptera treh orthologues are species specific. In aphids, genomic regions show lower synteny, which can be explained with chromosome holocentrism. Hemiptera TREH isoforms have higher diversity suggesting a subfunctionalization event in this taxon. Aphid trehalases act as a plant defences suppressor, but plants have probably evolved TREH inhibitors, leading to the wide treh gene family radiation described here for aphids. The role of heat shock proteins (hsp) in phenotypic plasticity has never been studied in aphids. We sequenced the hsp90 gene in 17 M. persicae lineages with different reproductive modes and winged morph production. We discovered that mutations altering hsp90 proteins were present in anholocyclic lineages only suggesting that hsp90 may buffer hidden mutations that disrupt cyclical parthenogenesis. The foraging gene is a unique example of a single gene underlying animal behaviour/phenotypic plasticity, so we have characterized a portion of the for gene in 12 M. persicae lineages and discovered that propensity to winged morphs production and for mutations are related, since frameshift mutations disrupt winged morphs production.
