Riassunto analitico
Il tumore al colon retto e il tumore ovarico sono due tra le maggiori cause di mortalità nel mondo. Il tumore al colon retto causa 610.000 morti all’anno e quello ovarico è la principale causa di morte per tumori ginecologici maligni. Il trattamento si basa sulla chemioterapia, utilizzando farmaci antimetaboliti come il 5-FU o la Capacitabina, che hanno come target la Timoidilato Sintasi umana (hTS). Questo enzima omodimerico consente la produzione de novo di Timidina, necessaria per la sintesi e la riparazione di DNA. Mentre questa attività catalitica è specifica della forma dimerica dell’enzima, quella monomerica è capace di legarsi al suo mRNA inibendo la traduzione della proteina stessa. Questi antimetaboliti stabilizzano la forma dimerica, riducendo la quantità di monomero e quindi causando la sovraespressione di hTS. Ciò può indurre la resistenza al farmaco o dare tossicità. L’obbiettivo di questo progetto è mettere a punto inibitori dell’interazione proteina-proteina (PPI) capaci di spostare l’equilibrio dimero-monomero verso la forma monomerica, inibendo l’attività catalitica della hTS pur mantenendo quella regolatoria. A questo fine, ho svolto le seguenti attività: 1. Sintesi di un inibitore dell’enzima hTS legato a sonde fluorescenti; 2. Valutazione della sua attività inibitoria sulle cellule tumorali (tumore ovarico e al colon retto); 3. Analisi dell’internalizzazione cellulare attraverso tecniche di Imaging. Ho sintetizzato sette inibitori dissociativi, uno di questi con un gruppo funzionale adatto a legare la sonda Fluoresceina (LC1296-FITC) e la BODIPY (LC1296-BODIPY), attraverso due differenti vie sintetiche. Successivamente, nella seconda parte del mio lavoro, ho analizzato con saggio MTT la tossicità dei due composti-sonda sulle linee cellulari di tumore al colon retto (HT29) e tumore ovarico (A2780) resistenti al 5-FU e analizzato l’internalization score, l’apoptosi e il ciclo cellulare di cellule trattate per 24 ore attraverso l’imaging flow cytometry. Inoltre, ho fatto studi di co-localizzazione con microscopia confocale. Gli stessi esperimenti sono stati fatti per il peptide LR che è stato legato alla sonda FITC. Sulla linea cellulare A2780, l'LC 1296-FITC (inibizione 24,66% a 75µM) ha mostrato un relativo aumento dell'IC50 rispetto al composto non legato. Tuttavia, il LC 1296-BODIPY ha mostrato una riduzione dell'attività. D'altra parte, su HT29, entrambi i composti LC 1296 (-FITC -BODIPY) hanno mostrato un'azione citotossica inferiore rispetto al composto non legato. Le due linee cellulari mostrano differenti livelli di internalizzazione del composto LC1296 e una localizzazione in strutture vescicolari. La linea cellulare HT29 mostra valori più alti di internalization score pur avendo internalizzato una quantità inferiore di farmaco. Entrambi i composti hanno inoltre un effetto sui processi di apoptosi e di ciclo cellulare. I risultati finali mostrano la riuscita sintesi di due inibitori TS fluorescenti, facilmente internalizzati all'interno delle linee cellulari di tumore ovarico e al colon retto. Questi mostrano una ridotta attività inibitoria nei confronti della proteina ricombinante e delle cellule tumorali rispetto al composto non legato, probabilmente dovuta alla diminuzione del legame con il dimero hTS o alla difficoltà di uscire dalle strutture vescicolari. Nel complesso, gli esperimenti hanno evidenziato il meccanismo di azione del composto LC1296 legato alle sonde e hanno supportato l'attività inibitoria hTS all'interno delle cellule tumorali. LC1296 risulta interessante al fine di sviluppare nuovi inibitori dissociativi. Riconoscimento. Abstract
Colorectal and ovarian cancer are two of the major causes of mortality worldwide, being colorectal cancer responsible for 610.000 deaths/year and ovarian cancer the leading cause of gynaecological malignancy deaths. Treatments are based on chemotherapy, using antimetabolites like 5-FU and Capecitabine, targeting the human Thymidylate synthase (hTS). This homodimeric enzyme provides de novo source of thymidylate required for DNA synthesis and repair. While hTS catalytic activity is specific of the dimeric form, monomers can bind to its mRNA repressing hTS protein translation. These antimetabolites stabilize the homodimeric form, decreasing monomer levels, causing hTS overexpression, which might mediate drug resistance and cellular toxicity.
My thesis work is part of a project having as a general aim the development of protein-protein interaction inhibitors (PPI) capable of shifting the dimer-monomer equilibrium towards the monomeric forms, thus halting the hTS catalytic function and preserving its regulatory activity. The specific aim is to characterize the cellular behaviour of thymidylate synthase inhibitors inside the cells. With this purpose, I developed the following activities: 1. Synthesis of a fluorescently labelled TS inhibitor; 2. Evaluation of the inhibitory potency of the labelled compound against the cancer cell (ovarian cancer and colorectal cancer); 3. Analysis of the inhibitor cellular internalization pathway via imaging techniques.
I synthesized seven dissociative inhibitors, one of them with specific functional group suitable for labelling with Fluorescein (LC1296-FITC) and BODIPY (LC1296- BODIPY), by a two ways-synthesis. Subsequently, in the second part of my work, I evaluated the two toxicity of the compounds by MTT assay on colorectal (HT29) and ovarian (A2780) cancer cells 5-FU resistant and analysed the internalization score, apoptosis and mitosis of the cells treated for 24 hours by imaging flow cytometry. Moreover, the co-localization was studied with the confocal microscopy. The same experiments were done for the LR peptide that was labelled with FITC.
On the A2780 cell line, the LC1296-FITC (inhibition 24.66% at 75µM) showed a relative increase on the IC50 compared to the unlabelled compound. However, the LC12976-BODIPY showed a more reduced activity. On the other hand, on HT29, both compounds LC1296-FITC and LC1296-BODIPY displayed a lower cytotoxic compared to the unlabelled compound.
The two cell lines showed different degree of LC1296 internalization and a cellular localization into vesicle-like structures. However, the HT29 cell line showed higher values of internalization score while displaying a lower total amount of the compound in the cell. They also show a relative effect on apoptosis and cell cycle.
The final results are related to the successful synthesis of two fluorescently labelled TS inhibitors, easily internalized inside colorectal and ovarian cancer cell lines. These compounds show a reduced inhibitory activity against the recombinant protein and cancer cells compared to the unlabelled compound, probably due to lower binding to the hTS dimer or its prevention from vesicle escape.
Overall, the experiments performed highlight the mechanism of action of labelled compound LC1296 and account for its hTS inhibitory activity inside the cancer cells.
LC1296 is an interesting compound with a novel mechanism of action that could be developed into more active inhibitors by lead optimization process.
