Riassunto analitico
Questa tesi ha come obiettivo la valutazione di un prototipo di imballaggio funzionale realizzato con materiali compostabili (poliidrossibutirrato-co-valerato, PHBV) e dotato di attività antimicrobica per inclusione di etil lauroil arginato (LAE). Il PHBV appartiene alla famiglia dei poliidrossialcanoati (PHAs), ovvero biopolimeri biodegradabili ottenuti da fonti rinnovabili attraverso bioprocessi, dotati di caratteristiche che li candidano a valide alternative agli imballaggi alimentari in plastica, per alcuni impieghi. La prima parte del lavoro sperimentale è stata dedicata alla determinazione della concentrazione minima inibitoria (MIC) del formulato commerciale Mirenat®-G verso muffe isolate da un campione di formaggio alterato. Successivamente, è stata valutata la resistenza termica dello stesso formulato, nell’ottica dell’eventuale incorporazione del principio attivo in matrici polimeriche mediante estrusione per la produzione di film e imballaggi attivi; nel dettaglio, si è voluta valutare la persistenza dell’efficacia antimicrobica del formulato commerciale dopo trattamento a diverse temperature (150-160-170°C), simulando l’eventuale danno termico prodotto durante le fasi di trasformazione dei polimeri. La seconda parte del lavoro ha avuto come obiettivo la valutazione dell’interazione tra imballaggio attivo e matrice alimentare attraverso la determinazione del livello di migrazione globale dal materiale verso simulanti alimentari svolta mediante prove di migrazione in conformità con il Regolamento (UE) 10/2011. Infine, con lo scopo di valutare l’efficacia del prototipo di active packaging nel rilasciare quantità di principio attivo tali da determinare un’inibizione dei microrganismi alteranti, sono stati effettuati dei test antifungini utilizzando aliquote del simulante posto a contatto con coppette del materiale attivo. I risultati dimostrano che trattamenti termici corrispondenti a condizioni di estrusione non comportano un’alterazione dell’efficacia antifungina del principio attivo e che, pertanto, è giustificata l’inclusione del LAE in matrici polimeriche per estrusione al fine della creazione di film e imballaggi attivi. Dall’altra parte, i prototipi testati hanno mostrato livelli di migrazione globale significativamente superiori ai limiti di legge, il che è da attribuire all’elevata concentrazione di glicerolo nel formulato commerciale: tale aspetto, che sicuramente limita lo scale-up commerciale per applicazioni alimentari, potrà essere oggetto di miglioramento attraverso la valutazione di formulati alternativi del principio attivo.

Abstract
The aim of this research thesis is the evaluation of a functional packaging prototype made by compostable materials (polyhydroxybutyrate-co-valerate PHBV) provided with antimicrobial activity because of the inclusion of ethyl lauroyl arginate (LAE). PHBV belongs to the family of polyhydroxyalkanoates (PHAs), which are biodegradable biopolymers obtained from renewable sources through bioprocesses, with properties that make them candidates for a valid alternative to plastic food packaging, for some applications.
The first part of the experimental work is focused on the determination of the minimum inhibitory concentration (MIC) of commercial formulation called Mirenat®-G, against molds isolated from a spoiled sample of cheese. Consequently, the thermal resistance of the same compound was evaluated in order to consider an eventual incorporation of the active ingredient in polymer matrix through extrusion for the production of films and active packaging. In detail, the objective was to examine the persistence of the antimicrobial effectiveness of the commercial formulation Mirenat®-G after exposure to different high temperatures (150-160-170°C), by simulating the eventual thermal damage produced during the transformation processes of polymers.
The second part of the work was based on the assessment of the interaction between the active packaging and the food matrix through the determination of the level of global migration from the material to the food simulants, carried out by means of migration tests according to the Regulation (UE) 10/2011. Furthermore, antifungal tests have been carried out using aliquots of the simulant after contact with cups made of active material, in order to evaluate the effectiveness of the active packaging prototype in releasing active ingredient quantities able to inhibit the spoilage microorganisms. The results show that thermal treatments, corresponding to extrusion conditions, do not implicate an alteration of the antifungal effectiveness of the active ingredient and, therefore, the inclusion of LAE in polymer matrix through extrusion for the production of films and active packaging is justified. On the other hand, the tested prototypes showed levels of global migration significantly higher than limits established by the law and that is attributed to the high concentration of glycerol in the commercial formulation: this aspect, that definitely represents a limit to the commercial scale-up for food application, can be improved through the evaluation of alternative formulation of the active principle.
