Al giorno d’oggi sono sempre di più i consumatori che ricercano alimenti salutari e in grado di tutelare la propria salute dai più svariati tipi di patologie; si pone molta più attenzione alle scelte alimentari che si compiono quotidianamente e si opta sempre di più per un’alimentazione consapevole. A tal proposito le aziende agro-alimentari si impegnano per mettere a punto metodologie e strumenti per aumentare quelle sostanze che hanno dimostrato avere un effetto positivo sulla salute umana, come i composti fenolici, per quindi soddisfare le esigenze dei consumatori. È ormai noto che una dieta ricca di polifenoli, metaboliti secondari presenti soprattutto negli alimenti di origine vegetale, svolga un’azione protettiva nei confronti di diverse patologie cronico-degenerative e malattie cardiovascolari. In particolare, gli agrumi, risultano essere ricchi di composti fenolici, tant’è che sono stati fino ad ora identificati più di sessanta flavonoidi presenti negli alimenti appartenenti al genere Citrus. Questi frutti rappresentano uno dei principali settori ortofrutticoli a livello mondiale in cui l’Italia si posiziona al tredicesimo posto per le esportazioni e al decimo per le importazioni. Gli agrumi, oltre ad essere destinati al consumo fresco, vengono utilizzati anche per produrre alimenti trasformati come succhi di frutta. In merito a questo, il protagonista di questo elaborato è stato un succo di arancia rossa della specie ‘Sanguinello’, le cui piante sono state trattate con fitoregolatori, in particolare brassinosteroidi e melatonina, per trenta giorni durante l’intero arco del loro sviluppo. L’utilizzo di queste sostanze in agricoltura è aumentato nel corso degli anni poiché risultano essere coinvolte in importanti processi metabolici come la crescita e lo sviluppo vegetativo delle piante (radicazione, senescenza fogliare, resa della biomassa ed efficienza fotosinetica), la maturazione di semi e frutti, nonché l’aumento della risposta a stress sia abiotici che biotici. Oltre a queste funzioni si è potuto constatare come tali composti potessero svolgere un ruolo rilevante anche nella sintesi di composti fenolici, influenzando direttamente l’espressione dell’enzima PAL, responsabile della sintesi di polifenoli. L’obiettivo di questa tesi è stato quello di verificare come brassinosteroidi e melatonina influenzassero il contenuto di composti fenolici presenti nel succo: attraverso l’utilizzo di HPLC è stato possibile identificare e quantificare flavonoidi e antocianine. Sono stati effettuati esperimenti anche per valutare l’attività antiossidante e alcuni parametri di qualità come pH, contenuto di solidi solubili totali (TSS), colore e fenoli totali. I risultati ottenuti, sottoposti poi ad elaborazione statistica, hanno mostrato come i trattamenti eseguiti con melatonina si siano rivelati particolarmente significativi sull’aumento del contenuto delle molecole in questione e come svolgano un ruolo degno di nota anche nell’attività antiossidante.

Abstract
Nowadays more and more consumers are looking for healthy foods, able to protect their health from different types of diseases; a lot more attention is paid to the food choices that are made every day and more and more people are opting for a conscious diet. In this regard, food businesses are committed to developing methods and tools to increase those substances that have been shown to have a positive effect on human health, such as phenolic compounds, to satisfy the needs of consumers. It is well known that a diet rich in polyphenols, secondary metabolites found especially in foods of vegetable origin performing a protective action against various chronic and degenerative pathologies such as cardiovascular diseases. In particular, citrus fruits are rich in phenolic compounds, and more than sixty flavonoids have been identified in foods belonging to the genus Citrus. These fruits represent one of the main fruit and vegetable sectors in the world in which Italy ranks in thirteenth place for exports and tenth for imports. Citrus fruits, besides being intended for fresh consumption, are also used to produce processed foods such as fruit juices. Concerning this, the protagonist of this paper was a red orange juice obtained from red orange fruit of the "Sanguinello" species, whose plants have been treated with phyto-regulators (brassinosteroids and melatonin) for thirty days during the entire time of their development. The use of these substances in agriculture has increased over the last years, as they are involved in important metabolic processes such as plant growth and vegetative development (rooting, leaf senescence, biomass yield and photosynthetic efficiency), seed and fruits maturation as well as increasing the response to both abiotic and biotic stresses. In addition to these functions, it has been observed that they could play an important role also in the synthesis of phenolic compounds, directly influencing the expression of the enzyme PAL, responsible for polyphenol synthesis. The objective of this thesis was therefore to verify if brassinosteroids and melatonin were able to affect the content of phenolic compounds present in the juice. The most important flavonoids and anthocyanins present in the red orange juices have been identified and quantified by means of HPLC analysis. Experiments were also carried out to evaluate the antioxidant activity and some quality parameters such as pH, total soluble solids content (TSS), color and total phenols. The results obtained, then subjected to statistical processing, showed that the treatments performed with melatonin were the most effective in increasing the flavonoids and anthocyanins content and the antioxidant activity of the red orange juices.
