Il phylum Tardigrada comprende circa 1300 specie adattate a diversi habitat, sia terrestri che acquatici. Il genere Xerobiotus (Eutardigrada, Macrobiotidae), il cui nome fa riferimento ad ambienti aridi come muschi su dune sabbiose o rocce esposte al sole in cui si trovano frequentemente tardigradi, è relativamente raro e poco studiato. Ad oggi, solo quattro specie sono state attribuite a questo genere: Xerobiotus pseudohufelandi Iharos, 1966 Xerobiotus xerophilus Dastych & Alberti, 1990 Xerobiotus euxinus Pilato et al., 2011 e Xerobiotus gretae Massa et al., 2021. In base a recenti dati molecolari di filogenesi è stata recentemente ipotizzata la soppressione del genere Xerobiotus e l'inserimento delle sue specie nel genere Macrobiotus, nonostante la presenza di caratteristiche morfologiche sinapomorfiche del genere sembrano al contrario sostenere la sua validità tassonomica. Infatti, per quanto è attualmente noto, nessun altro gruppo di tardigradi possiede un primo paio di zampe ridotto rispetto al secondo e al terzo, una struttura "tipo giarrettiera" che circonda le zampe, una diversa morfologia degli artigli tra l'ultima e le altre tre paia di zampe, l'assenza della lunula negli artigli delle prime tre paia e la presenza di pori cuticolari dorsolateralmente. Interrogativi sulla posizione filogenetica di questo taxon rimangono quindi aperti in attesa di ulteriori dati. Le analisi svolte per questa tesi hanno lo scopo di arricchire, attraverso l'utilizzo di un approccio morfologico e molecolare, le conoscenze relative alla biodiversità e biogeografia del genere Xerobiotus. Esemplari di tardigradi estratti da diversi campioni di muschio italiani ed esteri sono stati sottoposti a osservazioni al microscopio ottico ed elettronico e a caratterizzazione molecolare. Le informazioni ottenute sono state integrate da precedenti studi sul cariotipo e sulla modalità riproduttiva. Indagini mediante microscopia hanno mostrato, oltre alle strutture morfologiche distintive del taxon, caratteri nuovi o ancora poco noti per il genere Xerobiotus. Indagini molecolari, consistenti nel sequenziamento dei geni mitocondriali cox1 e cyt-b, i geni rRna 18S, 28S e ITS2, hanno permesso la caratterizzazione delle popolazioni e la costruzione di reti di aplotipi per gli individui analizzati. I dati indicano che un'ampia distribuzione geografica delle specie non è sempre correlata con le strategie riproduttive di partenogenesi. Questi studi chiariranno la controversa validità tassonomica del genere Xerobiotus e le variazioni morfologiche, molecolari e cariologiche presenti tra gli individui campionati in diverse regioni del globo, che potrebbero portare all'identificazione di specie non ancora note.

Abstract
The phylum Tardigrada includes about 1300 species adapted to different habitats, both terrestrial and aquatic. The genus Xerobiotus (Eutardigrada, Macrobiotidae), whose name refers to arid environments such as mosses on sandy dunes or rocks exposed to the sun in which tardigrades are frequently found, is relatively rare and little studied. To date, only four species have been attributed to this genus: Xerobiotus pseudohufelandi Ihaross, 1966 Xerobiotus xerophilus Dastych & Alberti, 1990 Xerobiotus euxinus Pilato et al., 2011 and Xerobiotus gretae Massa et al., 2021. Based on recent molecular phylogenetic data, the genus Xerobiotus has been hypothesized to be suppressed, and its species inserted in the genus Macrobiotus, despite the presence of synapomorphic morphological characteristics of the genus that on the contrary seem to support its taxonomic validity. Indeed, as far as is currently known, no other group of tardigrades possesses a reduced first pair of legs compared to the second and third, a "garter-like" structure that surrounds legs, a different morphology of the claws between the last and the other three pairs of legs, the absence of the lunula in the claws of the first three pairs of legs and the presence of cuticular pores dorsolaterally. Questions on the phylogenetic position of this taxon therefore remain open waiting for more data.
The analyses carried out for this thesis are aimed at enriching, through the use of a morphological and molecular approach, the knowledge related to biodiversity and biogeography of the genus Xerobiotus. Specimens of tardigrades extracted from different both Italian and foreign moss samples have been subjected to optical and electron scanning microscopy observations and molecular characterization. The informations obtained have been supplemented by previous karyotype and reproductive modality studies. Microscope investigations have shown, in addition to the distinctive morphological structures of the taxon, new or still little-known characters for the genus Xerobiotus. Molecular investigations, consisting in the sequencing of the mitochondrial genes cox1 and cyt-b, the genes rRna 18S, 28S and ITS2, have allowed the characterization of populations and the construction of networks of haplotypes for the individuals analysed. Data show that a wide geographical distribution of species is not always correlated with parthenogenetic reproductive strategies. These studies are going to clarify the debated taxon validity of the genus Xerobiotus and the morphological, molecular and cariological variations present between the individuals sampled in different regions of the globe, which could lead to the identification of species not yet known.
