Riassunto analitico
Le fenolamidi sono composti fenolici presenti in molte piante e costituiscono il prodotto della coniugazione di un acido fenolico con arilamine o poliamine. Essendo divenute oggetto di ricerca solo recentemente, in letteratura sono presenti poche informazioni riguardo attività, identificazione e separazione. I composti fenolici estratti da fonti naturali sono studiati soprattutto per la loro attività antiossidante, molto importante per la salute umana, in quanto correlata ad un minor rischio di insorgenza di patologie come cancro e malattie cardiovascolari. Il pomodoro è una considerevole fonte di sostanze antiossidanti nella dieta grazie alla presenza di molecole essenziali come il licopene, l’acido ascorbico, il tocoferolo e i composti fenolici. La pianta di pomodoro è stata scelta come fonte naturale per questo studio perché contiene diversi tipi di fenolamidi, che rappresentano l’oggetto principale della presente ricerca. I composti fenolici sono stati estratti dal frutto con estrazione solido-liquido, usando come solvente il metanolo nel primo metodo e l’etanolo nel secondo metodo, con l’utilizzo sia del pomodoro fresco che del pomodoro congelato. Lo studio analitico è stato condotto inizialmente con la spettrofotometria UV-VIS, per controllare la presenza di fenolamidi nell’estratto. Successivamente, la loro presenza è stata confermata dall’analisi HPLC, importante anche per la loro separazione. L’estratto fenolico è stato poi liofilizzato in modo da essere testato per l’attività antiossidante, in differenti concentrazioni. Sono stati eseguiti due diversi tipi di test antiossidanti: il saggio TBARS per la valutazione dell'inibizione della perossidazione lipidica e il saggio DPPH. Un’analisi completa richiederebbe tanto tempo, molto di più di quello dedicato a questo studio. Per questo, l’analisi dei risultati ottenuti è stata effettuata confrontandoli con valori presenti in altre ricerche, riproducendo simili condizioni sperimentali. Il presente studio su composti fenolici estratti dal pomodoro ha mostrato nel complesso risultati positivi sia riguardo l’analisi chimica sia riguardo i test sull’attività antiossidante. Innanzitutto, sono stati validati due differenti metodi di estrazione e sperimentate diverse condizioni di preparazione del campione per l’analisi e per i test. Sono state effettivamente individuate due molecole identificate con le fenolamidi, presenti nel frutto di questa pianta: caffeoil putrescina e tricumaroil spermidina; sono state applicate differenti condizioni per l’analisi HPLC che hanno permesso di individuare il metodo migliore per una più completa separazione dei picchi, elemento importante per un futuro isolamento di questi composti. Inoltre, è stata rilevata per i composti fenolici una buona attività antiossidante, studiata sia come inibizione della perossidazione lipidica che come attività anti-radicalica. Questi test hanno inoltre permesso di valutare gli effetti di differenti tipi di estrazione sull’attività antiossidante. Questa ricerca potrebbe costituire le basi di un futuro studio più approfondito in quest’area, incentrato sull’ottimizzazione delle tecniche di separazione e una migliore caratterizzazione della loro attività antiossidante.

Abstract
Phenolamides are phenolic compounds ubiquitous in several plants and they are products of the conjugation between a phenolic acid with arylamines or polyamines. As a recent object of research, poor information regarding their activity, identification and separation are present in literature. Phenolic compounds from natural sources are studied in major depth for their antioxidant activity, which is important for the human health because correlated with a lower risk of diseases such as cancer and cardiovascular complications. Tomato is a significant source of antioxidants in the diet, especially because its consumption is higher than all other fruits and vegetables and it includes essential molecules with this activity, such as lycopene, ascorbic acid, tocopherol and phenolic compounds.
The tomato plant was chosen as the natural source for this study because, according to literature, it includes some types of phenolamides, which represent the main object of the present research. In this work, phenolic compounds were extracted from the fruit with a solid-liquid extraction, using methanol for the first method and ethanol for the second method, using either the fresh fruit and the frozen fruit. The analytical investigation started with the analysis in spectrophotometry UV-VIS, to check the presence of phenolamides in the extract. Their presence was confirmed with HPLC, which allowed also their separation. Furthermore, the phenolic extract was lyophilized to be tested for its antioxidant activity, in different concentrations. Two types of antioxidant tests were performed: TBARS assay for evaluation of lipid anti-peroxidation activity and the DPPH radical scavenging assay.
A complete analysis requires a lot of time, much more than the dedicated to this study. For this reason, the analysis of the results was performed through comparison with values included in other studies, reproducing similar experimental conditions. This work on phenolic compounds extracted from tomato fruit produced positive results, both regarding chemical analysis and antioxidant tests. First, two different methods of extraction were validated and two different conditions of sample preparation for the analysis and tests were experimented. This allowed the detection of two molecules that can be identified as phenolamides in tomato fruit (caffeoyl putrescine and tricoumaroyl spermidine) and the different conditions for the HPLC analysis that were carried out allowed to choose the best method for a complete separation of peaks, which is important for a future isolation of these compounds. Furthermore, it was found that these compounds have a good antioxidant activity, both through inhibition of lipid peroxidation and radical scavenging mechanisms. These tests also allowed to evaluate the effect of the different types of extraction on the antioxidant activity of the extracts.
This research could constitute the basis of a more developed study about these compounds, focusing on the improvement of the separation techniques and further characterization of their antioxidant activity.
