Il genere Xerobiotus viene eretto nel 1996 da Bertolani e Biserov basandosi principalmente su caratteri fenotipici; ad oggi a questo genere sono state attribuite quattro specie: Xerobiotus pseudohufelandi Ihaross, 1966 Xerobiotus xerophilus Dastych & Alberti,1990 Xerobiotus euxinus Pilato et al., 2011 and Xerobiotus gretae Massa et al., 2021. Questo genere attualmente risulta oggetto di forte discussione in quanto mostra una notevole vicinanza genetica al genere Macrobiotus e per questo ne è stata anche ipotizzata la soppressione. Proprio quest’anno si è però dimostrata la presenza di sinapomorfie rilevanti che mettono in forte discussione la soppressione del genere: resta dunque un capitolo aperto, in attesa di maggiori dati, la filogenesi dello Xerobiotus. Ciò che invece appare evidente sono i caratteri sinapomorfici che differenziano il genere Xerobiotus da tutti gli altri, all’interno della famiglia Macrobiotidae. Nessun altro genere presenta infatti un primo paio di zampe ridotto rispetto al secondo ed al terzo, una struttura a “giarrettiera”che circonda le zampe ad aumentare l’attrito, una differente morfologia dell’unghia tra l’ultimo e le altre tre paia di zampe, l’assenza della lunula nell’unghia delle prime tre paia di zampe e la presenza di pori cuticolari dorsolateralmente. Tali adattamenti possono rappresentare la specificità di questo genere, eretto come gruppo di eutardigradi tipico di ambienti xerici, dunque caratterizzati da periodi di siccità o di aridità, come ambienti desertici o rocciosi.
In quest’ analisi, attraverso un approccio morfologico e molecolare, si è cercato sia di arricchire le banche dati in termini genetici ma anche fenotipici, riguardanti il genere Xerobiotus, sia di considerare la biodiversità e la biogeografia di specie di Xerobiotus rinvenute in diverse località. Per questa tesi sono stati raccolti sette campioni: dall’Italia (C-2859, C-3512, C-3518, C-3522,e C4446), dall’Olanda (C-3550) e dalla Georgia (C-3834). Quattro di questi campioni (C-2859, C-3512, C-3522, C-3834) hanno dimostrato una ricca popolazione del genere Xerobiotus e sono stati utili nell’ottenimento di risultati sia morfologici che molecolari standardizzabili. Per le osservazioni morfologiche, sono stati usati microscopio ottico e Scanning Electron Microscopies. Per le caratterizzazioni molecolari e gli studi filogenetici, i geni 18S, 28S, ITS-2 e CO1 sono stati sequenziati.
Dal punto di vista morfologico le quattro popolazioni si differenziano tra loro sia a livello di individui sia nelle uova; notevoli differenze sono riscontrabili anche all'interno delle stesse popolazioni. Dal punto di vista molecolare, la caretterizzazione molecolare attraverso la comparazione delle distanze genetiche della CO1 ha individuato un numero di gruppi di individui maggiore rispetto alle popolazioni analizzate.

Abstract
Xerobiotus genus was erected in 1996 by Bertolani and Biserov based mainly on phenotypic characters; to date, four species have been attributed to this genus: Xerobiotus pseudohufelandi Ihaross, 1966 Xerobiotus xerophilus Dastych & Alberti, 1990 Xerobiotus euxinus Pilato et al., 2011 and Xerobiotus gretae Massa et al., 2021.
This genus is currently the subject of strong discussion as it shows a considerable genetic proximity to the Macrobiotus genus and for this reason its suppression has also been hypothesized. Just this year, however, the presence of relevant synapomorphies has been demonstrated that strongly question the suppression of the genus. Therefore, the phylogeny of Xerobiotus remains an open question, waiting for more data. Instead, what appears evident are the synapomorphic characters that differentiate the genus Xerobiotus from all the others, within the Macrobiotidae family. In fact, no other genus has a reduced first pair of legs compared to the second and third, a "garter-like" structure that surrounds the legs to increase friction during locomotion a different morphology of the claws between the last and the other three pairs of legs, the absence of the lunula in the claws of the first three pairs of legs and the presence of cuticular pores dorsolaterally.
These adaptations can represent the specificity of this genus, erected as a group of eutardigrades typical of xeric environments, that are characterized by periods of drought or aridity.
In this study, through a morphological and molecular approach, an attempt was made both to enrich the phenotypic and genetic databases, concerning the genus Xerobiotus, and to consider the biodiversity and biogeography of Xerobiotus species found in different locations. Seven samples were collected for this thesis: from Italy (C-2859, C-3512, C-3518, C-3522, and C4446), from Holland (C-3550) and Georgia (C-3834). Four of these samples (C-2859, C-3512, C-3522, C-3834) presented a rich population of the genus Xerobiotus and were useful in obtaining both morphological and molecular standardizable results. For morphological observations, optical light microscopy and Scanning Electron Microscopy were used. For molecular characterizations and phylogenetic studies, the 18S, 28S, ITS-2 and CO1 genes were sequenced.
From the morphological point of view, the four populations differ from each other both at the level of individuals and in the eggs; clear differences can also be found within the same populations.
From the molecular point of view, the molecular characterization through the comparison of the CO1 genetic distances has identified a greater number of groups of individuals than the populations analyzed.
