NUP98, un gene coinvolto in traslocazioni cromosomiche che causano sindromi mielodisplastiche e forme di leucemia mieloide acuta (AML), è partner di fusione con più di 20 proteine diverse, tra le quali fattori di trascrizione della famiglia Hox. NUP98 codifica per una nucleoporina che media il trasporto di RNA e proteine attraverso la membrana nucleare. Inoltre, attraverso l’interazione con RAE1, un importante fattore di esporto per l’RNA dal nucleo, interviene nel controllo della segregazione cromosomica durante la mitosi. Sono state osservate diverse forme di fusione, oltre a quelle con i geni aventi l’omeodominio, tra cui forme chimeriche con proteine a funzione non nota e con fattori di trascrizione non contenenti l’homeobox. L’espressione esogena di oncoproteine di fusione che coinvolgono la porzione N-terminale di NUP98 è stata vista causare difetti nella struttura del fuso mitotico e una segregazione aberrante dei cromosomi, accompagnate da un’anticipata degradazione delle securine. I dati finora raccolti hanno mostrato che queste proteine di fusione causano instabilità cromosomica, verosimilmente predisponendo le cellule alla trasformazione tumorale. Inoltre, cellule primarie trasfettate transientemente o infettate con alcune di queste fusioni, tra cui NUP98-HOXD13, dopo un massimo di 48h dall’introduzione dell’oncogene, arrestano la crescita e vanno incontro a morte cellulare. Lo scopo del lavoro è stata la produzione di due diversi sistemi per l’espressione dell’oncoproteina NUP98-HOXD13, entrambi basati sull’utilizzo di vettori lenti virali, uno costitutivo e l’altro inducibile da tetraciclina. Il fine è stato quello di analizzarne gli effetti in cellule staminali/progenitrici emopoietiche nelle varie fasi del ciclo cellulare e al fine di ottenere livelli di espressione più vicini a quelli fisiologici. Sono stati quindi messi a punto due costrutti lentiviarli: uno contenente l’oncogene sotto un promotore forte costitutivo, l’altro sotto il controllo di un promotore inducibile dalla tetraciclina. I costrutti sono stati prima testati tramite trasfezione transiente in cellule HEK293 e, successivamente trasdotti in cellule HEK293 e in cellule staminali/progenitrici ematopoietiche CD34+ . Su queste cellule sono state effettuate analisi di Western blot, immunofluorescenza, e citofluorimetria a flusso per lo studio delle caratteristiche e del funzionamento del sistema d’espressione. I dati raccolti dimostrano che entrambi i sistemi funzionano, pur essendo osservabile nel sistema inducibile, un discreto livello di espressione dell’oncoproteina anche in assenza di induzione con tetraciclina. Le analisi mediante Western blot hanno inoltre confermato gli effetti biologici che l’oncogene NUP98-HOXD13 ha sulla stabilità delle securine, mentre l’osservazione di piastre metafasiche ha rilevato nelle cellule HEK293 infettate un sensibile aumento della percentuale di mitosi aberranti in seguito all’espressione dell’oncoproteina di fusione.

Abstract
Chromosomal translocations involving the Nup98 gene have been repeatedly implicated in acute myelogenous leukemia (AML) and, less commonly in myelodysplastic syndrome (MDS). These translocations generate chimeric fusion proteins, all of which have in common the N-terminal half of the Nup98 protein. The C-terminal partner in these chimeras can be a homeodomain transcription factor such as HOXD13. The Nup98 protein is a nucleoporin, a component of the nuclear pore complex (NPC), that mediates RNA and protein transport throughout the nuclear membrane. NUP98 interacts with RAE1, an important RNA nuclear export factor and regulates chromosomal segregation during mitosis. Overexpression of chimeric proteins involving Nup98 causes defects in the structure of the mitotic spindle and the consequent aberrant segregation of chromosomes, together with untimely degradation of securin. This genomic instability could predispose cells to oncogenic transformation.
The purpose of this work was the production of an expression system, based on lentiviral vectors, for the constitutive or tetracycline inducible expression of the oncoprotein NUP98-HOXD13 in order to analyze its effects in hematopoietic stem/progenitor cells.
Two lentiviral construct were generated, one with the expression of the gene under the control of a strong promoter, the other under the control of a tetracycline-regulated promoter.
First of all, we tested the vectors by transient transfection in HEK293 cells, and then we transduced these cells for the generation of a stable cell line expressing the oncogene. Subsequently the fusion protein was expressed in CD34+ hematopoietic stem/progenitor cells to study its biological effects.
On transfected cells initially, and then on infected cells, we performed Western blot, immunofluorescence and flow citometry analysis to study the efficiency of the two systems.
The results of transient transfection showed that both systems work, even if the inducible system shows some leakiness in expression. In infected stable HEK293 cell line, Western blot analysis have confirmed the biological effects of the fusion protein on securin degradation, moreover chromosome count in metaphase spreads has shown a significant increase in the percentage of aberrant mitosis in NUP98-HOXD13 expressing cells.
