Riassunto analitico
Il tumore del Colon retto è una delle principali cause di mortalità in tutto il mondo ed è la terza forma di tumore più comune, responsabile di 610.000 decessi ogni anno. Il trattamento si basa principalmente sulla chemioterapia che utilizza come farmaci di prima linea il 5-FU, la Capecitabina e il Pemetrexed, antitumorali che interagiscono con l’enzima Timidilato sintasi (hTS), enzima indispensabile per la sintesi del DNA. Lo sviluppo di resistenza nel tumore del colon-retto è associato all’accumulo di mutazioni genetiche, tra cui una sovra-espressione dell’hTS. L’hTS svolge la sua funzione catalitica in forma dimerica, mentre la sua forma monomerica libera è cataliticamente inattiva ed è responsabile dell’azione di autoregolazione ossia dell’espressione dell’enzima stesso. Lo scopo di questo progetto è di indurre una perturbazione nell’equilibrio dimero-monomero a favore della forma monomerica, per inibire l’azione catalitica mantenendo quella regolatoria. Lo scopo della mia tesi di laurea è quello di sviluppare degli inibitori dissociativi in grado di perturbare l’equilibrio dimero-monomero. Con questo obiettivo, mi sono occupata della progettazione, sintesi, e valutazione dell’inibizione enzimatica e cellulare di nuovi composti potenzialmente dotati di meccanismo dissociativo. Nella prima parte del mio progetto di tesi attraverso un approccio misto di ligand-based e structure-based, ho provveduto alla progettazione e sintesi di 18 nuovi derivati, analoghi del lead compound (LC1321) ottenuto da studi precedenti, con l’intento di migliore le capacità dissociative-inibitorie e le caratteristiche farmacologiche. Il meccanismo di inibizione di questi composti è stato valutato mediante cinetica enzimatica e l’effetto dissociativo attraverso la metodica FRET. La maggior parte dei composti riporta buoni valori di variazione di FRET in un range che varia tra -0,101 e -0,624 rispetto al valore massimo osservabile in condizioni non dissociative (per il dimero) pari a 1, attestandone la capacità di destabilizzare la forma dimerica dell’enzima. I composti 1g e 3c presentano valori di IC50 rispettivamente pari a 15 μM e 5 μM e inducono un calo nell’efficienza di FRET rispettivamente pari a -0.624 e a -0.446 a 40μM. La capacità di 1g e 3c di inibire l’hTS, associata a buoni valori di FRET, supporta un meccanismo di azione per tali composti di tipo dissociativo. Successivamente, nella seconda fase del mio progetto di tesi, ho provveduto a valutare l’attività di inibizione della crescita delle cellule tumorali dei composti sintetizzati attraverso saggi di citotossicità cellulare su 3 linee cellulari di tumore colon-retto (CRC), resistenti al 5-FU, e 2 linee cellulari di carcinoma ovarico (OC), resistenti al cisplatino. La maggior parte dei composti induce una morte cellulare pari al 20-30% a 40μM, dipendentemente dalla linea cellulare considerata. In particolar modo, i due composti A21 e C5 mostrano una percentuale di morte cellulare comparabile a quella del composto di riferimento (LC1343), confermando il valore dello scaffold chimico di partenza. Il composto A21 ha un’attività citotossica pari al 56% sulla linea di OC e al 59% sulla linea di CRC, mentre il composto C5 ha un’attività pari al 67% sulla linea di CRC e al 52% sulla linea di OC, entrambi alla concentrazione di 40μM. Sulla base dei dati ottenuti è stato quindi possibile identificare nuovi promettenti composti, quali agenti terapeutici in grado di agire come inibitori dissociativi della hTS, con la peculiarità di evitare la sovraespressione della proteina bersaglio. Il mio contributo personale a questo lavoro di tesi ha riguardato la sintesi di 18 composti e la valutazione citotossica cellulare di 32 composti. Questo progetto di tesi si inserisce all’interno del progetto AIRC2015-IG16977 ‘Protein-protein interaction inhibitors of thymidylate synthase against colorectal cancer’.

Abstract
Colorectal cancer (CRC) is one of the major causes of mortality throughout the world and it is the third most common form of cancer with 610.000 deaths per year.
The treatment is mainly based on chemotherapy, employing 5-FU, Capecitabine and Pemetrexed as first line drugs, antimetabolites which interact with hTS (human Thymidilate synthase) enzyme, a key protein involved in the synthesis of DNA. In colorectal cancer, the development of resistances is associated with accumulation of genetic mutations, such as hTS overexpression. hTS plays its catalytic function in homodimeric form, whereas the free monomer is catalytically inactive and it autoregulates the expression of hTS itself.
The aim of this project is to induce a perturbation in the dimer-monomer equilibrium in favour of the monomeric form, in order to inhibit the catalytic function preserving the regulatory activity.
The goal of my experimental degree thesis is to develop new dissociative inhibitors able to perturb the dimer-monomer equilibrium. With this purpose, I dealt with the design, synthesis and evaluation of the enzymatic and cellular inhibitory activity of new compounds, potentially endowed by dissociative mechanism.
In the first part of my thesis project, through a mixed ligand-based and structure-based approach, I focused on the design and synthesis of 18 new derivatives, analogues of a lead compound (LC1321) obtained from previous studies, with the intent to improve the dissociative-inhibitor activity and the pharmacological features. The inhibition mechanism for these compounds was studied through enzymatic kinetic assays and the dissociative effect through FRET methodologies. Most of the compounds showed good FRET values in a range between -0,101 and -0,624 at 40μM in comparison with the maximal observable value equal to 1 in non-dissociative condition (for the dimeric form), attesting the capability of these compounds to disrupt the dimeric form of the enzyme. 1g and 3c reported hTS inhibitory IC50 of 15 μM and 5 μM respectively and they induced a drop in FRET efficiency respectively equal to -0.624 and to -0.446 at 40μM. The capability of 1g and 3c to inhibit the hTS, associated with good FRET values, support a mechanism of action based on a dissociative inhibition.
Subsequently, in a second phase of my work, I evaluated the cancer cell grow inhibitory activity of all the synthesized compounds through cytotoxicity assays against three CRC, 5-FU resistant, and two OC (ovarian cancer), cisplatinum resistant, cell lines.
The majority of the compounds induced a cellular death around 20-30% at 40 μM, related to cell line considered.
In particularly, A21 and C5 showed a percentage of cell death comparable to the reference compound (LC1343), supporting the importance of the starting chemical scaffold. Evaluating the cytotoxicity activity against OC and CRC cell line respectively, for A21 56% and 59% has been observed while for C5 resulted in 67% and 51%, both at the concentration of 40μM.
Basing on these results, it was possible to identify new promising compounds, representing novel therapeutic agents acting as dissociative inhibitors of hTS, with the peculiarity to avoid target protein overexpression.
My own contribution to this thesis work was focused on the synthesis of 18 compounds and cytotoxicity cellular evaluation of 32 compounds.
