Riassunto analitico
Questo progetto è stato eseguito nel laboratorio della professoressa Gertrud Morlock, presso il dipartimento di scienze nutrizionali dell’università Justus Liebig di Giessen (Germania), grazie al programma Erasmus +, coordinato dalla professoressa Federica Pellati. Lo scopo di questa tesi era ottenere i profili biologici di 10 campioni di canapa, mediante effect-directed analysis (EDA). Dopo l’estrazione dei 10 campioni, in questa ricerca è stato usato il metodo HPTLC-UV-Vis-EDA-ESI-HRMS, che consiste in un’analisi cromatografica ad alta prestazione, seguita da effect-directed analysis utilizzando derivatizzazioni microchimiche, saggi biologici (Aliivibrio fischeri e Bacillus subtilis) ed enzimatici (α e β-glucosidasi, tirosinasi, acetilcolinesterasi e p-YES); alla fine le zone che hanno mostrato l’attività biologica maggiore sono state caratterizzate con la spettrometria di massa (MS). Per mezzo di analisi MS, è stato possibile identificare nei campioni analizzati la presenza di acido cannabidiolico (CBDA) e acido cannabidivarinico (CBDVA), che sono i composti più coinvolti nell’attività biologica riscontrata: questi composti hanno mostrato attività antibatterica contro Aliivibrio fischeri e Bacillus subtilis, e attività di inibizione enzimatica verso gli enzimi α e β-glucosidasi, tirosinasi e acetilcolinesterasi. CBDA ha mostrato anche attività antiossidante, valutata utilizzando la derivatizzazione microchimica con il reagente DPPH (radicale 2,2-difenil-1-picrilidrazile). Il metodo utilizzato è stato molto efficiente, rapido e sensibile, e potrebbe essere utile in molti campi, soprattutto per analisi farmaceutiche e alimentari.

Abstract
This project was carried out in the laboratory of Prof. Dr. Gertrud Morlock, Department of Nutritional Science (Justus Liebig University, Giessen, Germany), thanks to the Erasmus + Program, coordinated by professor Federica Pellati. The aim of this thesis work was to obtain a bio-profiling of ten hemp samples, through effect-directed analysis (EDA). After the extraction of the samples, the method used in this research was HPTLC-UV-Vis-EDA-ESI-HRMS, which consists of high-performance chromatographic analysis, followed by effect-directed analysis using microchemical derivatization, bioassays (Aliivibrio fischeri and Bacillus subtilis) and enzymatic assays (α and β-glucosidase, tyrosinase, acetylcholinesterase and p-YES). Then, the zones that showed the strongest bioactivity were characterized using mass spectrometry (MS). By means of MS, it was possible to identify in the samples analysed, the presence of cannabidiolic acid (CBDA) and cannabidivarinic acid (CBDVA), which are the two most involved compounds in the biological activity observed: these compounds showed antimicrobial activity against Aliivibrio fischeri and Bacillus subtilis, and also enzymatic inhibition towards the enzymes α and β-glucosidase, tyrosinase and acetylcholinesterase. CBDA showed antioxidant activity as well, evaluated using the microchemical derivatization with the reagent DPPH (2,2-diphenyl-1-picrylhydrazyl radical). The method used proved to be very efficient, rapid and sensitive, and it could be useful in many fields, involving mainly pharmaceutical and food analysis.
