La lattuga è una delle colture a foglia più cultivate nel mondo. Il fenomeno del bolting in lattuga corrisponde alla fioritura e causa una riduzione del valore della coltura, la quale è economicamente fruibile solo fino a prima di tale stadio di sviluppo. Il bolting causa quindi una perdita economica e i breeder cercano di ridurre al minimo questo fenomeno. Il bolting è indotto da ormoni vegetali (es. gibberelline), fattori ambientali (es. temperatura, ore di luce) e stress nutrizionali. Meccanismi simili sono osservati anche in altre colture come cipolla, canna da zucchero, girasole o arabidopsis. La genome wide association analysis (GWA) valuta la diversità di una popolazione al fine di identificare polimorfismi correlati con la variazione fenotipica. GWA è un processo che richiede diversi passaggi: selezione di una collezione di linee, fenotipizzazione del tratti di interesse, genotipizzazione high-throughput, analisi della struttura della popolazione e associazione tra marcatori e fenotipi. E’ stata costituita una collezione di 153 genotipi di lattuga, collezionate tra linee di breeding, accessioni pubbliche, cultivar commerciali e linee di interesse per la ricerca. La collezione è stata fenotipizzata per i tratti correlati al bolting (ritardo del bolting e velocità di bolting) in 4 ambienti, differenti per anno, stagione e località, e genotipizzata con un approccio di genotyping by sequencing (GBS). Più di 20000 SNPs sono stati ottenuti e posizionati sul genoma di lattuga. Con l’ausilio del software STRUCURE e dell’analisi delle componenti principali (PCO) è stata analizzata la strutturazione della popolazione. I dati suggeriscono la presenza di 5 sotto popolazioni, le quali tendono a sovrapporsi con le tipologie di lattughe presenti nella collezione (romane, foglie di quercia, cappuccio, batavie e crisphead). I dati ottenuti sono stati utilizzati per applicare i modelli GLM e MLM+Q+K tramite il software TASSEL e trovare SNPs associati con il bolting. Le sequenze contenenti gli SNPs si trovano principalmente su LG7 e LG9 e correlano con proteine coinvolte nel meccanismo di bolting in lattuga e in altre specie.

Abstract
Lettuce is one of the most cultivated leafy vegetable crop in the world. Bolting is the flowering of lettuce and cause a reduction of economic value of the crop which is usually limited to a pre-flowering stage. In this respect bolting causes a relevant economic loss and the breeders work to reduce bolting as much as possible. Bolting is triggered by plant hormones (e.g. gibberellins), environmental factors (e.g. day length, temperatures) and nutritional stresses. Similar mechanisms has been observed in other crops like onion, sugarcane, sunflower or Arabidopsis.
Genome wide association analysis evaluates the genetic diversity of a population in order to identify polymorphism correlated with phenotypic variations. Association analysis is a process encompassing different steps: selection of a panel of germplasm, phenotypization of the traits of interest, genotypization of the germplasm with high-through put molecular markers, analysis of the population structure and association between markers and phenotypes.
A collection of 153 genotypes of lettuce was generated collecting lines used in breeding programs, ancestors frequently observed in the pedigrees of popular cultivars and commercial varieties. The panel was phenotyped for bolting-related traits (Bolting Delay and Bolting Speed) in 4 environments, differing for year, season and location and genotyped with a Genotype by Sequencing (GBS) method.
More than 20000 SNPs were obtained and located on reference lettuce genome. The analysis of the population structure of the association panel was conducted with STRUCTURE software and PCO approach. The data suggest the presence of 5 sub-populations overlapped with the typologies of lettuce present in collection (romaine, oakleaf, butterhead, crisphead, batavia). Obtained data were used to apply GLM and MLM+Q+K statistical method with TASSEL software in order to identify SNPs associated with bolting. Several sequences correlated with proteins involved in bolting mechanism in lettuce and other species were mainly found in LG7 and LG9.
