EfTS: un nuovo target per lo sviluppo di farmaci antiinfettivi PhD:Debora Cortesi Le Infezioni causate da Enterobacter Faecalis rappresentano un problema importante per la salute causato dalla resistenza ai farmaci.
Abbiamo estratto la timidilato sintasi (TS) da questo microrganismo , potenziale bersaglio per la terapia antibiotica. La TS (CE: 2.1.1.45) catalizza la metilazione riduttiva di 2'deossiuridina-5'-monofosfato (Dump) a 2'-deossitimidina-5'-monofosfato(dTMP), con il cofattore N5-N10 metilentetraidrofolato (MTHF) .4,8,9 . La TS rappresenta l'unica fonte di dTMP in cellule umane ed è un bersaglio importante per la progettazione di farmaci chemioterapici. Tra gli antitumorali noti sono utilizzati in terapia 5-fluorouracile e Raltitrexed. Abbiamo identificato una libreria di derivati ftalimidici: 2-metil-1H-isoindolo-1,3 (2H) -dione N- derivati e (2-metil-1,3-diosso-2,3-diidro-1H-isoindol-4-il) metil] ammino derivati (JMedChem2011) che hanno mostrato attività inibitoria verso TS batteriche, compresa la TS di Enterobacter faecalis (EfTS). Per allargare la classe degli inibitori di EfTS abbiamo continuato con un approccio diretto basato sul ligando verso l’individuazione di inibitori di EfTS attraverso il quale si potrebbero ottenere più numerosi composti affini. Abbiamo selezionato una libreria di 150 composti ftalimidici. Tra questi 53 sono stati selezionati in base a criteri di diversità strutturale, peso molecolare e solubilità (logP). La libreria selezionata è stata testata verso TS di Lactobacillus casei TS (LcTS) e TS umane (hTS). I composti sono stati selezionati in base alla loro attività e specie-specificità verso LcTS, EcTS, e hTS. LcTS, EcTS e hTS sono stati purificati. Gli esperimenti di cinetica enzimatica sono stati condotti attraverso test cromogenici in condizioni standard. La classe di composti che abbiamo analizzato ha evidenziato una debole inibizione verso EfTS. Tuttavia di oltre 56 composti che abbiamo testato, quasi la metà hanno mostrato di inibire le proteine (12). In particolare,è stato analizzato l’inibitore SS7, perché la sua struttura cristallina dimostra che è legato solo ad una subunità di ogni dimero EfTS allo stesso modo come 5F-THF (un debole inibitore della proteina). Il complesso EFTs-5_FTHF subisce un cambiamento conformazionale che rappresenta una limitata accessibilità al sito di legame per il d-UMP (11). I modelli di inibizione dei composti sono stati determinati mediante analisi cinetica allo stato stazionario della dipendenza dell'attività enzimatica sulla concentrazione MTHF al variare delle concentrazioni di inibitore. Tutti i composti hanno mostrato inibizione competitiva verso MTHF.

Abstract
EfTS: a new target for anti-infective drug development

Abstract
Infections caused by Enterobacter Faecalis represent an important health problem for therapy
caused by drug resistance.
We targeted Tymydylate Synthase (TS) from this organism as a potential target for
antibacterial therapy. TS (EC: 2.1.1.45) catalyzes the reductive methylation of 2’-
deoxyuridine-5’-monophosphate (dUMP) to 2’-deoxythymidine-5’-monophosphate
(dTMP), assisted by the cofactor N5, N10-methylene tetrahydrofolate (MTHF).4,8,9 Since
TS represents the only synthetic source of dTMP in human cells, it is a major target for the
design of chemotherapeutic agents.
10 Among the known drugs, 5-Fluorouracil and Raltitrexed are in therapy as anticancer agent. We
have identified a library of phtalimide derivatives, 2-methyl-1H-isoindole-1,3(2H)-dione N-
derivatives (JMedChem2011) that showed inhibitory activity against bacterial TS including Enterobacter
faecalis TS (EfTS).
To enlarge the EfTS inhibitor class we continued the ligand based approach directed towards the
identification of EfTS inhibitors that could be translated in highest affinity compounds. We
started from the phthalimidic scaffold (1H-isoindole-1,3(2H)-dione ) and selected a new library
based on N-substituted phthalimidic derivatives.
We selected anew library of 150 phtalimidic compounds. Among them 53 were selected based
on criteria of structural diversity, molecular weight and solubility (logP). The selected library
was tested against the TS enzyme biolibrary including EfTS, Lactobacillus casei TS (LcTS) and
human TS (hTS). The compounds were screened for their activity and their species-specificity
against LcTS, EcTS, EfTS, and hTS. LcTS, EcTS, and hTS were purified .
Enzyme kinetics experiments were conducted by chromogenic assay under standard
conditions.
The compound class we have analysed has proven weakly inhibiting the EfTS protein .
However over 56 compounds we have tested, almost the half showed to inhibit at some level the
EfTS protein(12).
In particular, SS7 inhibitor was analysed, because it’s crystal structure shows that it is bound
only in one subunit of each EfTS dimer in the same way as 5F-THF(a weak inhibitor of the
protein).
The complex EfTS-5_FTHF undergoes a conformational changes that accounts for the limited
accessibility to the d-UMP binding site(11).
The inhibition patterns of the compounds were determined by steady-state kinetic analysis
of the dependence of enzyme activity on MTHF concentration at varying inhibitor
concentrations.
All the compounds showed competitive inhibition with respect to MTHF.
