In questa tesi sono stati studiati gli effetti benefici di batteri lattici (LAB) isolati da alimenti carnei fermentati, con particolare attenzione alla loro capacità di inibire lo sviluppo di batteri contaminanti degli alimenti ed alla potenziale capacità di esercitare un’azione probiotica. Inoltre sono state utilizzate moderne tecniche di analisi delle popolazioni batteriche complesse per studiare la popolazione di bifidobatteri nelle feci di infanti allattati al seno da madri classificate secretor (FUT2+) e non-secretor in base alla capacità di produrre 2’-fucosil-lattosio (2FL), al fine di valutare l’effetto prebiotico degli oligosaccaridi del latte materno (HMO). Relativamente agli effetti benefici dei LAB, è stata valutata la loro potenziale applicazione come biopreservanti. Infatti i LAB possono produrre batteriocine, peptidi ad azione antibatterica nei confronti di L. monocytogenes, E. coli, E. faecalis e S. aureus. I LAB sono stati isolati da insaccati fermentati e sono stati studiati per la capacità di inibire i suddetti patogeni. Un ceppo di Lactobacillus delbrueckii ha mostrato una marcata attività anti Listeria ed è stato utilizzato con successo in un challenge test in cui è stato condotto il processo di fermentazione della carne. Inoltre è stato valutato se i LAB presenti nei prodotti carnei e/o utilizzati come starter nella produzione di salumi potessero sopravvivere al transito gastrointestinale e potessero raggiungere il colon, un prerequisito affinché i batteri possano esercitare un’azione probiotica. La sopravvivenza dei batteri nello stomaco dipende dalla loro capacità di tollerare i pH bassi. La tolleranza alla bile è invece attribuita alla produzione di bile salt hydolase. Ceppi appartenenti a lattobacilli normalmente utilizzati come starter per la produzione insaccati fermentati (soprattutto L. sakei) sono stati sottoposti a screening per la loro resistenza all’acidità e ai sali biliari. La maggior parte di essi ha tollerato valori di pH di 3.5, ma nessuno ha tollerato i sali biliari. Quindi, nonostante i batteri analizzati possano essere efficaci nel contenere la contaminazione batterica dell’alimento, la loro sopravvivenza al transito gastrointestinale e quindi la loro capacità di esercitare un’azione probiotica in vivo, sembra improbabile. A differenza dei LAB ottenuti dagli alimenti fermentati, una solida dimostrazione dell’effetto probiotico è disponibile per i bifidobatteri. Questi sono commensali naturali dell’intestino, che colonizzano già nei primi giorni di vita, specialmente nei neonati allattati al seno, anche se le dinamiche della colonizzazione non sono ancora state chiarite. In questo studio, è stata confrontata la popolazione di bifidobatteri nelle feci di neonati alimentati con latte di madri secretor o non-secretor. Per analizzare la popolazione dei bifidobatteri e determinarne la tassonomia sono state utilizzate la biotipizzazione mediante MALDI-TOF MS, l’analisi del polimorfismo dei frammenti terminali di digestione (Bif-TRFLP) di amplificati PCR del gene 16S, ed un saggio PCR per l’identificazione delle sottospecie di B. longum (longum o infantis). I bifidobatteri isolati dagli infanti di madri FUT2+ erano dominati dalle sottospecie del gruppo B. longum, mentre quelli isolati da infanti di madri non-secretor erano dominati da B. breve. La capacità degli isolati di fermentare il 2FL è stata studiata, dimostrando che le specie di Bifidobacterium nelle feci dei neonati di madri FUT2+ riflettono l’adattamento all’utilizzo di questo HMO. I dati presentati dimostrano che il 2FL è un oligosaccaride prebiotico che esercita un’efficace azione bifidogenica nei confronti di alcune specie di Bifidobacterium.

Abstract
In this thesis, the beneficial effects of LAB isolated from fermented meat products were studied, with particular focus on their ability to inhibit food-borne pathogens and on their potential ability to function as probiotics. Furthermore, modern molecular techniques for the investigation of complex mixed bacterial communities were utilized to study the population of bifidobacteria in the feces of breast-fed infants receiving milk from secretor (FUT2+) and non-secretor mothers, based on the ability to produce 2’-fucosyllactose (2FL), to evaluate the prebiotic effect of the human milk oligosaccharides (HMO). With respect to the beneficial effects of LAB, their potential utilization as food bioprotectants was first investigated. In fact, some LAB can secrete bacteriocins, antibacterial peptides against L. monocytogenes, E. coli, E. faecalis, or S. aureus. LAB were isolated from fermented sausages and screened for their ability to inhibit these pathogens. A strain of Lactobacillus delbrueckii exhibiting a marked anti Listeria activity and was identified and successfully utilized for challenge test in a meat fermentation process. Furthermore, it was investigated whether the LAB occurring in meat products and/or specifically selected to guide meat fermentation may survive the transit through the upper gastrointestinal tract and may reach the colon, a prerequisite for bacteria to exhibit any probiotic property. The survival of bacteria in gastric juice depends on their ability to tolerate low pH value. Bile tolerance of bacteria has been suggested to be associated to bile salt hydrolase. Several strains belonging to Lactobacillus species normally utilized as starters for meat fermentation (especially L. sakei) were screened for their resistance to high acidity and high concentration of bile components. Most of the strains resisted a pH value of 3.5, but none of them survived the bile salts. Therefore, despite some strains of LAB can restrict food-borne pathogens in fermented products, it seems improbable that they could reach the colon to exert any probiotic function in vivo. Unlike LAB from fermented foods, solid demonstration of the probiotic function and the health benefits of bifidobacteria have been provided in the last decades. Bifidobacteria are normal inhabitants of the human hindgut, which is colonized in the very first days after birth, especially in breast-fed newborns even if the dynamics of bifidobacterial colonization of infants remain to be clarified. In this thesis, the bifidobacterial population in the feces of newborns receiving breast milk from secretor (FUT2+) and non-secretor mothers was compared. To analyze bifidobacterial population and to determine the taxonomy of bifidobacterial species, MALDI-TOF MS biotyper, terminal restriction fragment length polymorphism (Bif-TRFLP) on PCR amplicons of 16S genes, and PCR-based assay for determination of B. longum subspecies (subsp. longum vs infantis) were utilized. Fecal isolates from infants that receiving milk from FUT2+ mothers were dominated by B. longum group species, while the ones from infants that received milk from non-secretor mothers were dominated by B. breve species. The bifidobacterial isolates were screened for the ability to ferment 2FL, thus demonstrating that the relative composition of Bifidobacterium species in feces of infants receiving milk from secretor mothers reflects the adaptation to the utilization of this HMO.
The development of the new techniques and methods contributes to our understanding of the dynamics within the intestinal microbiota and enables a thorough investigation of the population of probiotic and/or commensal bifidobacteria.
